Iron As Therapeutic Targets In Human Diseases

Iron is an essential element for almost all organisms, a cofactor playing a crucial role in a number of vital functions, including oxygen transport, DNA synthesis, and respiration. However, its ability to exchange electrons renders excess iron potentially toxic, since it is capable of catalyzing the formation of highly poisonous free radicals. As a consequence, iron homeostasis is tightly controlled by sophisticated mechanisms that have been partially elucidated. Because of its biological importance, numerous disorders have been recently linked to the deregulation of iron homeostasis, which include not only the typical disorders of iron overload and deficiency but also cancer and neurodegenerative diseases. This leads iron metabolism to become an interesting therapeutic target for novel pharmacological treatments against these diseases. Several therapies are currently under development for hematological disorders, while other are being considered for different pathologies. The therapeutic targeting under study includes the hepcidin/ferroportin axis for the regulation of systemic iron homeostasis, complex cytosolic machineries for the regulation of the intracellular iron status and its association with oxidative damage, and reagents exploiting proteins of iron metabolism such as ferritin and transferrin receptor. A promising potential target is a recently described form of programmed cell death named ferroptosis, in which the role of iron is essential but not completely clarified. This Special Issue has the aim to summarize the state-of-the-art, and the latest findings published in the iron field, as well as to elucidate future directions.

The pleasant community of Limone suI Garda provided outstanding hospitality for a second NATO ARW dealing with apolipoprotein variants, which are natures clues for the discovery of the physiological roles of apolipoproteins in lipoprotein metabolism in normal subjects and patients with specific dyslipoproteinemias. Limone, the site of discovery of the first human apolipoprotein mutant, apoA-I-Milano, provided a brilliant sunny spring venue for more than 50 participants from both sides of the ocean. The attendance at the colorful opening ceremony of the ARW was one of the largest on record. Two members of the Italian government, the Secretaries of Health and the Navy, gave the welcoming addresses. Six television networks, two with national audiences, covered the international workshop. The Limone oracles provided a montage of insights gleamed from the eyes of the clinican, the biochemist, and the molecular biologist. The cumulative information on the molecular defects in lipoprotein metabolism reviewed by this diverse group of investigators provided an ever expanding horizon of new knowledge in this fast moving and some times perplexing field. Clinical vignettes were presented on patients from throughout the world including Canada (Connelly), Turkey (Schmitz), and France (Infante) detailing the clinical sequelae of a defect in a specific apolipoprotein. The clinical importance of Lp(a), a lipoprotein relegated almost to obscurity for many years, has now taken v center stage.

This publication of a symposium held on 24 th and 25 th of June 1988 in Munich th is dedicated to Nepomuk Zollner on the occasion of his 65 birthday, expressing the best wishes of the authors. Nepomuk Zollner was born in the northern part of Bavaria. While a medical student in Munich he was called up to military duty in the last year of World War II. After achieving excellent results on his examinations he served as a physician in several hospitals in Munich and soon became interested in inborn errors of metabolism. In order to receive the best education possible at that time he joined the group of S. J. Thannhauser, the famous German emigrant, in Boston where he worked from 1951 to 1953. There he was able to continue his studies on lipids, which he had started in 1948, and begin with his studies on purine metabolism. While working in Thannhauser's laboratory he learned to think and act according to the strict laws of natural SCIence. After returning to Europe he concentrated his scientific work on purines and lipids. Gout and hyperlipoproteinemias from the genetic to the therapeutic aspects remained the dominating topics of his research activities. In spite of many obligations as head of the Medical Polyclinic of the University of Munich and many activities at the university and in scientific societies Zollner's foremost intent continued to be the development and progress of natural science in medicine.
