Targeting The Dna Damage Response For Anti Cancer Therapy

Over the past decade a complex role for DNA damage response (DDR) in tumorigenesis has emerged. A proficient DDR has been shown to be a primary cause for cellular resistance to the very many DNA damaging drugs, and IR, that are widely used as standard-of-care across multiple cancer types. It has also been shown that defects in this network, predominantly within the ATM mediated signaling pathway, are commonly observed in cancers and may be a primary event during tumorigenesis. Such defects may promote a genomically unstable environment, facilitating the persistence of mutations, any of which may provide a growth or survival advantage to the developing tumor. In addition, these somatic defects provide opportunities to exploit a reliance on remaining repair pathways for survival, a process which has been termed synthetic lethality. As a result of all these observations there has been a great interest in targeting the DDR to provide anti-cancer agents that may have benefit as monotherapy in cancers with high background DNA damage levels or as a means to increase the efficacy of DNA damaging drugs and IR. In this book we will review a series of important topics that are of great interest to a broad range of academic, industrial and clinical researchers, including the basic science of the DDR, its role in tumorigenesis and in dictating response to DNA damaging drugs and IR. Additionally, we will focus on the several proteins that have been targeted in attempts to provide drug candidates, each of which appear to have quite distinct profiles and could represent very different opportunities to provide patient benefit.

Information Highways are widely considered as the next generation of high speed communication systems. These highways will be based on emerging Broadband Integrated Services Digital Networks (B-ISDN), which - at least in principle - are envisioned to support not only all the kinds of networking applications known today but also future applications which are not as yet understood fully or even anticipated. Thus, B-ISDNs release networking processes from the limitations which the communications medium has imposed historically. The operational generality stems from the versatility of Asynchronous Transfer Mode (ATM) which is the transfer mode adopted by ITU-T for broadband public ISDN as well as wide area private ISDN. A transfer mode which provides the transmission, multiplexing and switching core that lies at the foundations of a communication network. ATM is designed to integrate existing and future voice, audio, image and data services. Moreover, ATM aims to minimise the complexity of switching and buffer management, to optimise intermediate node processing and buffering and to bound transmission delays. These design objectives are met at high transmission speeds by keeping the basic unit of ATM transmission - the ATM cell - short and of fixed length.

Feynman Motives

This book presents recent and ongoing research work aimed at understanding the mysterious relation between the computations of Feynman integrals in perturbative quantum field theory and the theory of motives of algebraic varieties and their periods. One of the main questions in the field is understanding when the residues of Feynman integrals in perturbative quantum field theory evaluate to periods of mixed Tate motives. The question originates from the occurrence of multiple zeta values in Feynman integrals calculations observed by Broadhurst and Kreimer.Two different approaches to the subject are described. The first, a ?bottom-up? approach, constructs explicit algebraic varieties and periods from Feynman graphs and parametric Feynman integrals. This approach, which grew out of work of Bloch?Esnault?Kreimer and was more recently developed in joint work of Paolo Aluffi and the author, leads to algebro-geometric and motivic versions of the Feynman rules of quantum field theory and concentrates on explicit constructions of motives and classes in the Grothendieck ring of varieties associated to Feynman integrals. While the varieties obtained in this way can be arbitrarily complicated as motives, the part of the cohomology that is involved in the Feynman integral computation might still be of the special mixed Tate kind. A second, ?top-down? approach to the problem, developed in the work of Alain Connes and the author, consists of comparing a Tannakian category constructed out of the data of renormalization of perturbative scalar field theories, obtained in the form of a Riemann?Hilbert correspondence, with Tannakian categories of mixed Tate motives. The book draws connections between these two approaches and gives an overview of other ongoing directions of research in the field, outlining the many connections of perturbative quantum field theory and renormalization to motives, singularity theory, Hodge structures, arithmetic geometry, supermanifolds, algebraic and non-commutative geometry.The text is aimed at researchers in mathematical physics, high energy physics, number theory and algebraic geometry. Partly based on lecture notes for a graduate course given by the author at Caltech in the fall of 2008, it can also be used by graduate students interested in working in this area.
