BOOK EXCERPT:
Gene expression is an active ongoing process that maintains a functional CNS, as proteins are being made on a continual basis. Processes such as learning and memory, nerve cell repair and regeneration and its response to stress are critically dependent on gene expression. This volume highlights the role of gene expression in normal CNS function, and presents many research methods at the cutting edge of neuroscience, which will provide insight into therapeutic approaches through which the control of gene expression may be used in the treatment of many nervous system diseases.

BOOK EXCERPT:
~he major theme of this book is the development of the vertebrate central nervous system. ~is volume contains summaries of most of the invited participants at the NA~ advanced study institute entitled "Development of central nervous system in vertebrates" held in Maratea, Italy, from June 23-July 5, 1991. In order to address this topic, we have drawn upon a selection of current studies dealing with molecular, cellular and system analysis which specifically pertain to the general principles of the development. ~he major aim of this institute was to bring together a select group of investigators who would present their views on the current issues in their respective fields and to foster extensive discussions amongst participants in smaller groups. Such interactions brought together the exchanges of ideas amongst participants and helped clarify the intricate details and formulate new vistas and collaborations. Since the study of nervous system development has focused mostly on the origin of neuron and glia cells, the area of current research was represented by talks on early cellular events including effects of growth factors, BOX and other gene expressions and cell lineage of specific cell type(s). Formation of specific cell types and the specific neuronal connections have been a major theme in the study of the nervous system development. Recent technical advances has resulted in new information at both cellular and molecular levels which have provided new details. Current research was represented by "selective" topics discussed at the meeting.

BOOK EXCERPT:
Prominent experimentalists critically review the animal models widely used in developing powerful new therapies for central nervous system diseases. Coverage includes novel uses of animal models of Alzheimer's, Parkinson's, and Huntington's diseases, and studies of aging. Techniques that rely heavily on behavioral analyses, as well as models developed from infusions of neurotoxins and from advances in molecular biology, are thoroughly explicated, as are models developed for more acute neurological conditions, including traumatic brain injury and stroke. Comprehensive and authoritative, Central Nervous System Diseases: Innovative Animal Models from Lab to Clinic offers neuroscientists, pharmacologists, and interested clinicians a unique survey of the most productive animal models of the leading neurological diseases currently employed to develop today's innovative drug therapies.

BOOK EXCERPT:
Few areas of biomedical research provide greater opportunities to capitalize upon the revolution in genomics and molecular biology than gene therapy. This is particularly true for the brain and nervous system, where gene transfer has become a key technology for basic research and has recently been translated to human therapy in several landmark clinical trials. Gene Therapy in the Brain: From Bench to Bedside represents the definitive volume on this subject. Edited by two pioneers of neurological gene therapy, this volume contains contributions by leaders who helped to create the field as well as those who are expanding the promise of gene therapy for the future of basic and clinical neuroscience. Drawing upon this extensive collective experience, this book provides clear and informative reviews on a variety of subjects which would be of interest to anyone who is currently using or contemplating exploring gene therapy for neurobiological applications. Basic gene transfer technologies are discussed, with particular emphases upon novel vehicles, immunological issues and the role of gene therapy in stem cells. Numerous research applications are reviewed, particularly in complex fields such as behavioral neurobiology. Several preclinical areas are also covered which are likely to translate into clinical studies in the near future, including epilepsy, pain and amyotrophic lateral sclerosis. Among the most exciting advances in recent years has been the use of neurological gene therapy in human clinical trials, including Parkinson's disease, Canavan disease and Batten disease. Finally, readers will find "insider" information on technological and regulatory issues which can often limit effective translation of even the most promising idea into clinical use. This work provides up-to-date information and key insights into those gene therapy issues which are important to both scientists and clinicians focusing upon the brain and central nervous system.

BOOK EXCERPT:
Immediate-early genes are believed to be involved in the neuron's ability to con vert short-term synaptic stimulation into long-lasting responses and thus contribute to the adaptive alterations involved in neuronal plasticity. Cellular immediate-early genes share a close structural homology with some viral oncogenes. Recent advances in cellular biology have identified the activation and deactivation of immediate-early genes as molecular mechanisms to control regulated and deregulated growth, cellular differentiation and development. In this view immediate-early genes may function as third messengers in a stimulus transcription cascade transferring extracellular information into changes in target gene transcription, thereby changing the phenotype of neurons. Immediate-Early Genes in the Central Nervous System provides a comprehensive up-to-date overview of current methodology in the research of immediate-early genes and includes a wide range of neurobiological topics, such as regeneration, memory formation, epilepsia and nociception. The contributors to this book have been selected from among the leading experts in their field of research. T.R. TOLLE J. SCHADRACK W. BOOK EXCERPT:
This book presents the reader with an understanding of the role played by matrix metalloproteinases (MMPs) in the normal and diseased central nervous system (CNS). These enzymes may be important to brain development, and may also contribute to tissue destruction, which is observed with inflammatory and degenerative conditions of the brain. The book provides a background on the biology of MMPs, and on the stimuli and conditions that are linked to an increase in their production and activity. It describes the targets of MMPs, which include matrix proteins such as collagen, soluble cytokines and chemokines, and cell surface receptors. Studies implicating MMPs in neuronal process outgrowth and cell migration in CNS development are covered. The book also touches on studies suggesting that, in certain situations, dysregulated MMP activity and/or production may be critical to blood-brain barrier breakdown and neuronal damage./a
