In recent years, the importance of the tissue surrounding a tumour, commonly termed the tumour microenvironment (TME), has been increasingly recognized. Whilst likely to play a tumour-suppressive role in the early stages of carcinogenesis, a wide array of evidence indicates the TME becomes corrupted by the presence of malignant cells and becomes tumour permissive, promoting growth, metastasis, immune evasion and therapeutic resistance. The major components of the TME are fibroblasts, immune cells, endothelial cells and the extracellular matrix in which they are embedded, but this is highly heterogeneous and site-specific, with other cells such as adipose, bone and neuronal likely to play a prominent role in some tumours. Other patient-specific factors are also likely to be important, including age and previous treatment. The multi-faceted contribution of the TME to tumourigenesis offers great potential for therapeutic intervention and biomarker development, but its inherent heterogeneity necessitates in-depth understanding of the contribution of different components of the TME to tumourigenesis to underpin clinical translation. This Research Topic will encompass a wide range of areas relating to the therapeutic opportunities offered by the tumour microenvironment of oral cancer, including (but not limited to) the role of fibroblasts, infiltrating immune and inflammatory cells and the extracellular matrix, including the influence of aging and previous treatment. The collection can encompass a range of oral tumours including mucosal cancers, salivary gland cancers and tumours of the jaws, including odontogenic tumours. This Research Topic will have a translational focus to it and may include biomarker development. Antimicrobial peptides and complement are distinct components of the innate immune defence. While antimicrobial peptides, after cleavage of a preproprotein, have the ability to insert directly in non host membranes, complement requires a sequential enzymatic activation in the fluid phase in order to produce a transmembrane membrane attack complex. Its insertion is controlled by membrane bound regulators. Deficiencies are described for both effectors and relate to increased susceptibility of infection. In addition, however, antimicrobial peptides and complement each influence the activity of inflammatory cells as recent data in the respective research areas shows. This series of articles draws together for the entities of antimicrobial peptides and complement a balance of contributions in the areas of evolution, roles, functions and preclinical applications. By comparing and contrasting antimicrobial peptides and complement, greater cross-disciplinary appreciation will be derived for their individual and overlapping spectra of activity, circumstances of activation and their general ability to more completely inform the inflammatory and cellular response.

A link between inflammation and cancer has been established many years ago, yet it is only recently that the potential significance of this connection has become apparent. Although several examples of chronic inflammatory conditions, often induced by persistent irritation and/or infection, developing into cancer have been known for some time, there has been a notable resistance to contemplate the possibility that this association may apply in a causative way to other cancers. Examples for such progression from chronic inflammation to cancer are colon carcinoma developing with increased frequency in patients with ulcerative colitis, and the increased incidence of bladder cancer in patients suffering from chronic Schistosoma infection. Inflammation and cancer have been recognized to be linked in another context for many years, i.e., with regards to pathologies resembling chronic lacerations or 'wounds that do not heal.' More recently, the immunology of wound healing has given us clues as to the mechanistic link between inflammation and cancer, in as much as wounds and chronic inflammation turn off local cell-mediated immune responses and switch on growth factor release as well the growth of new blood vessels - angiogenesis. Both of these are features of most types of tumours, which suggest that tumours may require an immunologically shielded milieu and a growth factor-rich environment.
