BOOK EXCERPT:

The dark side of the genome represents vast domains of the genome that are not encoding for proteins – the basic bricks of cellular structure and metabolism. Up to 98% of the human genome is non-coding and produces so-called long non-coding RNA. Some of these non-coding RNA play fundamental roles in cellular identity, cell development and cancer progression. They are now widely studied in many organisms to understand their function. This book reviews this expanding field of research and present the broad functional diversities of those molecules and their putative fundamental and therapeutic roles and develops the recent history of non-coding RNA, their very much debated classification and how they raise a formidable interest for developmental and tumorigenesis biology. Using classical examples and an extensive bibliography, the book illustrates the most studied and attractive examples of these long non-coding RNA, how they interface with epigenetics, genome integrity and expression and what are the current models of their regulatory mechanisms. - This book offers a large review about the long non-coding RNA - It presents the broad functional diversities of those molecules - It presents pioneer works from the field - Provides a comprehensive review of the field - Presents fundamental and therapeutic interests

Long Non Coding Rna

eBook Download

BOOK EXCERPT:

Long non-coding RNAs (LncRNAs) were originally regarded as “noise” in the genome due to their lack of protein-encoding capacity. However, their roles are now understood to cover various biological functions like gene regulation, cell proliferation, survival, differentiation, and chromatin remodeling. As an emerging group of non-coding RNA molecules, LncRNAs are involved in diverse diseases like cancer, cardiovascular, autoimmune, hormonal disorders, neurodegeneration, and infection, among others. However, knowledge of the mechanisms by which they act is still ambiguous. As such, this book meets the essential need for a textbook dedicated to this topic and provides a comprehensive and captivating account of lncRNA research.

Molecular Biology Of Long Non Coding Rnas

eBook Download

BOOK EXCERPT:

Long non-coding RNAs (lnc)RNAs have emerged as a new paradigm in epigenetic regulation of the genome. Thousands of lncRNAs have been identified and observed in a wide range of organisms. Unlike mRNA, lncRNA have no protein-coding capacity. So, while their function is not entirely clear, they may serve as key organizers of protein complexes that allow for higher order regulatory events. Discovering these functions has been the result of intense research done of the last few years, and lncRNA research has had several critical developments during that time. This book will consolidate these ideas and models to better examine the most important issues in lncRNA biology. This will include critical studies that have led to the discovery and annotation of lncRNAs in numerous species, and the molecular mechanisms for a few lncRNA that have begun to emerge.

BOOK EXCERPT:

Noncoding RNAs (ncRNAs) are the “dark matter” of the genome, occupying vast regions yet not coding for proteins. Despite their elusive nature, they play a pivotal role in regulating gene expression and cellular processes and participate in chromatin remodeling and signal transduction. Crucial in development and disease, ncRNAs influence embryogenesis, cell differentiation, and cancer progression. Their potential in diagnostics and therapeutics marks a revolutionary frontier in genetic and medical research. This book provides a comprehensive overview of the regulatory and developmental roles of ncRNAs, from gene regulation in plants to their crucial functions in embryogenesis and cancer, and their potential in diagnostics and therapeutics.

Noncoding Rna In Disease Diagnosis Etiology Progression Prognosis And Treatment

eBook Download

BOOK EXCERPT:

An exciting class of regulatory small noncoding RNAs (ncRNAs) has emerged in recent years that play key role(s) in the etiology/pathogenesis and diagnosis/prognosis of various human diseases including various hematologic and solid cancers, cardiovascular diseases including stroke/atherosclerosis, diabetes, obesity, and metabolic disorders, neuropathies, neurodegeneration including Alzheimer’s Disease, Parkinson’s Disease or Multiple Sclerosis, neurological disorders including epilepsy and muscular dystrophies, liver diseases, pulmonary fibrosis, immune-mediated inflammatory disorders and inflammatory bowel disease, gastritis, autoimmune disorders including arthritis, acromegaly, chronic kidney disease, allergic rhinitis, vitiligo, osteoporosis, obstructive sleep apnea, ocular disease, periodontitis, skeletal muscle atrophy, and parasitic diseases including leishmaniasis, amongst others. These ncRNAs include primarily microRNAs (miRNAs), long noncoding RNAs (lncRNAs), circular RNAs (circRNAs), piwi-interacting RNAs (piRNAs), small nucleolar RNAs (snoRNAs), small interfering RNAs (siRNAs) or exosomal RNAs (exRNAs), etc. that modulate the expression of target genes thus potentially regulating protein expression in normal versus diseased individuals. This Research Topic recognizes the importance of and seeks to explore, ncRNAs in the diagnosis, etiology, progression, prognosis, and treatment of human disease as well as novel biomarker discovery by multi-omics technologies and other methodologies. Indeed, high-throughput technologies employing deep sequencing, proteogenomic, microarrays, and quantitative real-time PCR have begun to shed new light on the architecture of ncRNA landscapes in various diseases and key signal transduction cascades modulated by these ncRNAs in disease pathology. Epigenetic control of gene expression mediated via miRNAs, for instance, has expanded our understanding of transcriptional and post-transcriptional regulation of gene expression by critical ncRNAs in the cellular milieu. Network biology approaches including the construction of relevant ncRNA networks, ncRNA-mRNA networks, and bioinformatics for mining of ncRNAs, are gaining greater relevance in the search for ‘target hubs’ or novel biomarker(s) of disease. Delineation of ncRNA signatures by transcriptomic profiling or RNA-seq, for example, is an important milestone in furthering our knowledge of the repertoire of specific ncRNAs implicated in various diseases. Further, elucidation of the specific steps of the signaling cascades where ncRNAs exert their regulatory actions will augment our understanding of diseases. This will lead to the development of (i) a novel biomarker, or biomarker signature panel, for disease; (ii) ncRNA-based companion diagnostics for diagnosis of disease; (iii) point-of-care testing devices based on ncRNAs; (iv) newer therapeutic agents that target novel biomarkers with specificity, thereby reducing toxicity; (v) ncRNA-based prophylactic approaches of the future; and, (vi) ncRNA-based prognosis of disease.
