Fungal Wilt Diseases of Plants focuses on wilt diseases caused by the fungal genera Verticillium, Fusarium, and Ceratocystis. Special attention is given to the interactions of physiological, biochemical, and anatomical factors, as these relate to pathogenesis and mechanisms of disease resistance. Organized into 16 chapters, this book begins with a description, in a historical perspective, of the major research themes in fungal wilt diseases. It then looks into the worldwide status of this plant disease. The three subsequent chapters describe the epidemiology and life cycle of the major fungal wilt pathogens in Fusarium, Verticillium, and Ceratocystis. This book also provides an in-depth view of the genetics and biochemistry of these pathogens; the nature of pathogenesis and the effects of wilt pathogens on host-water relations; and the sources and genetics of host resistance in field and fruit crops, vegetable crops, and shade trees. Other chapters are dedicated to the biochemistry, physiology, and the anatomical aspects of resistance and to the progress in the biological and chemical control of these pathogens. This text will be of great value to graduate students and senior research scientists in plant pathology, physiology, and biochemistry, who are specifically involved in studying wilt diseases and host-parasite interactions. It will provide them the detailed background information needed to supplement their specialized research interests.

Methods in Virology, Volume V focuses on the methods used in virology, including hybridization, gel electrophoresis, freeze-etching technique, and ultracentrifugation. The selection first offers information on the fusion of cells for virus studies and production of cell hybrids; approaches to ultracentrifugation; and polyacrylamide gel electrophoresis of viral RNA. Discussions focus on applications to virological problems and analysis of results; analysis of the distribution of RNA on polyacrylamide gels after electrophoresis; and biological analysis of DNA components. The book then examines the polyacrylamide gel electrophoresis of viral proteins, DNA-RNA and DNA-DNA hybridization in virus research, and techniques of RNA-DNA hybridization in solution for the study of viral transcription. Topics include preparation of nucleic acids, hybridization and elution procedures that minimize RNA degradation, and procedures for acrylamide gel electrophoresis. The text takes a look at freeze-etching technique for the study of virus ultrastructure; procedures to increase virus yield from infected plants; and the immunoperoxidase technique. Concerns include principles of the immunoperoxidase technique, histochemical detection of peroxidase activity, sequence of events in virus infection, and factors affecting virus yield. The selection is a valuable source of data for researchers interested in the methods employed in virology.

The Second Edition of this bestseller brings together basic plant pathology methods published in diverse and often abstract publications. The Second Edition is updated and expanded with numerous new figures, new culture media, and additional methods for working with a greater number of organisms. Methods are easy to use and eliminate the need to seek out original articles. This reference allows for easy identification of methods appropriate for specific problems and facilities. Scientific names of pathogens and some of their hosts are updated in this edition. The book also acts as a research source providing more than 1,800 literature citations. The Second Edition includes chapters on the following: Sterilization of culture apparatus and culture media Culture of pathogens with detailed techniques for 61 fungi and selected bacteria Long-term storage of plant pathogens Detection and estimation of inoculum for 28 soilborne fungal pathogens and 5 bacterial genera-15 methods for airborne inoculum and 13 methods for seedborne pathogens Establishment of disease and testing for disease resistance Work with soil microorganisms Fungicide evaluation Biological control Bright-field microscopy

Phytopathogenic Prokaryotes, Volume 1, provides an understanding of the diversity and complexity of diseases caused by phytopathogenic prokaryotes. It is part of a two-volume treatise that summarizes current research on these organisms. The book is organized into four parts. Part I covers the physical nature of prokaryotic phytopathogens as well as how they are presently classified, the limitation of this artificial classification, and the biology of the pathogen's invasion of plants. Part II presents conceptual hypotheses for the formation of the agricorpus (pathogen/host complex as a biological unit) and how this association may be detrimental or beneficial to both members of the unit. Other topics include the basic determinant of evolutionary change (the gene), and the evolution of vectors for dispersal of pathogens. Part III elaborates on the interaction at the plant/environment/pathogen interface (the plant surface). It presents information on the interaction of prokaryotes in the rhizosphere and phyllosphere, and how this interaction developed. Part IV shows how prokaryotes affect their hosts once infection has been established. This information is presented in sequence progressing from the disease-causing mechanisms of the facultative endophytic pathogens to those of the obligate endophytic pathogens.
